The passive drag (Dp) of 218 competitive swimmers was studied and related to their performance level. To study this relationship, specific attention was given to anthropometric and joint laxity (JL) variations. The Dp was measured at 1.40 m.s-1, using a mechanical winch and a strain gauge with a load cell connected to a strain bridge. Swimmers were towed in a prone position holding their breath after a maximal inspiration. Buoyancy was evaluated by the hydrostatic lift (HL), i.e., the maximal weight just necessary to maintain the swimmer in a balanced position under the water after a maximal inspiration. The JL was assessed by a standard scoring system. The Dp was related mainly to the surface area (SA) (r = 0.73 and 0.53; P less than 0.01, for males and females, respectively). For a given SA, Dp was inversely related to the performance level. The JL explained 7% of the variability of Dp. On average, Dp measured after a maximal expiration, increases of about 22% SD 3% (P less than 0.01). This increase was related to individual vital capacities (r = 0.86, P less than 0.01). As Dp was mainly related to SA and HL, it is suggested that the body exerts a large pressure effect on the water. The contribution to performance might be related to the gliding phase of swimming. 相似文献

The purpose of this study was to determine the relationship between blood gases and acid-base measurements in arterial, arterialized venous, and venous blood measured simultaneously during short-term maximal exercise. Ten well-trained male cyclists performed a graded maximal exercise test on a cycle ergometer to determine the power output corresponding to their peak oxygen consumption (test I), and a short-term maximal test on a cycle ergometer at peak power output (test II). During test II arterial, arterialized venous and venous blood were sampled simultaneously for determination of partial pressures of oxygen and carbon dioxide, pH, bicarbonate (HCO3-), base excess (BE), and lactate (La). Samples were taken at rest, the end of 1 min of exercise (1 ME), at the end of exercise (EE), and at 2 min of recovery (REC). During test II, subjects maintained a peak power output of 370.6 (62.1) W [mean (SD)] for 4.5, SD 1.6 min. Except at rest venous and arterialized venous measurements tended to be the same at all sampling intervals, but differed significantly from measurements in arterial blood (P less than 0.05). BE was the only variable that rendered consistently significant correlations between arterial and arterialized venous blood at each sampling interval. The pooled correlation coefficient between arterial and arterialized venous BE was r = 0.83 [regression equation: BEa = (0.84 BEav)-0.51]. Human lung fibroblasts (MRC5) were treated chronically with the oxygen radical-generating system hypoxanthine plus xanthine oxidase and assayed for malignant transformation in the soft agar colony assay. After a thrice weekly exposure to oxygen radicals for 4 and 5 weeks, there was a significant number of transformants compared to controls. In 4 separate experiments, karyotypes of the malignant transformants were examined. 22/75 metaphases exhibited karyotypic abnormalities and in 13/22 of the abnormal karyotypes, a t16:18 (p13.3,q21) translocation was observed. Methoxatin is a novel o-quinone coenzyme found in a variety of dehydrogenases and oxidases. In this paper we present the visible absorption spectra, apparent pKa, and midpoint potentials of the methoxatin and two analogous phenanthrolinedione semiquinones. We have also obtained absorption spectra for the semiquinone forms of the methoxatin-containing holoenzyme methylamine dehydrogenase and of its resolved methoxatin-containing subunit. The ornithine decarboxylase (ODC)-deficient Chinese hamster ovary (CHO) cell line C55.7 has normal amounts of ODC mRNA with very low amounts of immunologically detectable ODC protein, suggesting a structural mutation; however, 5-azacytidine treatment leads to phenotypical reversion (Steglich, C., and Scheffler, I. E. (1985) Somat. Cell Mol. Genet. 11, 11-23). We have demonstrated by chemical cleavage a single base mismatch in DNA heteroduplexes composed of wild-type and mutant cDNA strands. DNA sequencing showed that the mutant phenotype results from an aspartate-glycine substitution at amino acid 381 of the protein. When 5-azacytidine-revertant cell lines were selected for resistance to alpha-difluoromethylornithine, the resulting amplified ODC gene was structurally indistinguishable from the wild type gene. These results suggested the existence of a single active ODC locus in CHO cells. Using the methylation-sensitive restriction endonucleases AvaI and HpaII, we found evidence for two differentially methylated alleles in wild type, ODC-deficient and alpha-difluoromethylornithine-resistant cells. One of the alleles appeared completely inactivated by hypermethylation but could be reactivated by demethylation in spontaneous or 5-azacytidine-induced revertants. 相似文献

The uncI gene, the first gene of the unc operon, has been cloned into an expression vector carrying the lambda PRPL promoters in tandem orientation and the gene cI857 coding for the thermolabile repressor. Linkage of the uncI gene to an efficient ribosome binding site (the translational initiation region of the uncE gene) resulted in 10-20-fold increased gene expression. The i protein has been extracted from overproducing cells using chloroform/methanol and purified to homogeneity by ion exchange chromatography. Analyzing the products of the uncI gene encoded by different plasmids, we provide evidence that, in contrast to the previously reported data (Walker, J. E., Saraste, M., and Gay, N. J. A novel rat hepatocidal test, based on morphological changes in monolayer culture and the percentage of lactate dehydrogenase (LDH) released into the medium after exposure to culture filtrates of Listeria spp. was used to determine listerial toxicity and pathogenicity. Primary cultures of rat hepatocytes exposed to brain heart infusion (BHI) culture filtrates from ATCC strains of Listeria monocytogenes and L. ivanovii, released 91-92% and 95% of LDH after 3 h and 18.5 h, respectively. Cultured monolayers changed from normal hepatocytes into nonviable round forms. Brain heart infusion broth and BHI culture filtrates of other Listeria spp. were nontoxic to hepatocytes. The rat hepatocidal test is a quantitative and rapid system for studying listerial toxicity and pathogenicity. 相似文献
